Etymology: "Bestly" is a plain English formation: the superlative "best" with the adverbial suffix "-ly", so it means "in the best way." Its base is one of the most irregular words in English. "Best" descends from Old English "betst," the superlative of a root that also gave "better," and the two forms are historically unrelated to the positive "good." English speakers borrowed the suppletive pattern from the old root, which explains why the comparative and superlative look so unlike the base. Dictionaries record "bestly" as rare and mostly archaic, and it appears far less often than "best" itself or the common adverb "well." The suffix does the work here: English has long attached "-ly" to adjectives to form adverbs, and "best" followed the same route as "fair" or "kind."
